3 S A L U K I C L U B O f A M E R I C A Page 3 PRESIDENT S MESSAGE Winter 2016 Newsletter Another Holiday Season is upon us and the 2016 SCOA National Specialty is less than six months away. Planning and preparation are in process and I want to thank chair Monica Stoner and her committees in advance. I am confident that it will the same success as Dagmar Hintzenberg-Freisleben will be judging the regular classes and Shereen Shuman will be judging the sweepstakes classes. The Board received one proposal for the 2017 SCOA National Specialty which the Board approved. Russell McFadden will be the chair and it will be at Purina Farms. Because of a scheduling conflict at Purina, the date will be one week later than currently. It will take place on June 13 to 17, Kathy Webb will be judging the regular classes. All of SCOA s committees are in place and I want to thank the many volunteers for their work. On a sad note, we note the passing of Lin Hawkyard. SCOA has lost a dedicated and hard worker and all of us have lost a friend. We will try to notify the membership of members recently passing in our Newsletters, recognizing that there will be gaps because of imperfect information. Passing members are recognized in our National Specialty catalog each year and we will be sending a catalog to the families of those included. As always, if you have thoughts, ideas or questions, the Board welcomes comments to secretary@salukiclub. org. Present were Gary Roush, Linda Scanlon, Jennifer Napier, Debbie Feliciano, Katie Souder, Brian Duggan, Pam Arwood, Catherine Comroe, Sharon Furlong, Wanda Sirianni, Monica Stoner (Delegate) 1. Sharon Furlong moved to approve the minutes from the May 21, 2015 teleconference sent on June 22, Debbie Feliciano seconded the motion which passed with Linda Scanlon, Pam Arwood and Wanda Sirianni abstaining. 2. Katie Souder moved to approve the May financial reports sent on June 14, Linda Scanlon seconded the motion which carried unanimously. 3. Katie Souder moved to approve Junior Membership for Elizabeth Hodge Smith. Linda Scanlon seconded the motion which carried with Pam Arwood abstaining. 4. Debbie Feliciano moved to include the current judges list on the website as requested at the AGM. Sharon Furlong seconded the motion which carried unanimously. 5. Sharon Furlong moved to set the posting privileges on the breeder referral Facebook page to administrative only in accordance with the motion which was passed at the AGM. Catherine Comroe seconded the motion which carried unanimously. 6. There was a discussion regarding the concerns brought up at the AGM that it was difficult to find judges on SCOA s list that didn t judge a fair amount in the East. The suggestion was made that the options should include a longer list. Following the discussion Brian Duggan moved to increase the list of eligible judges to 30 listed alphabetically. Jennifer Napier seconded the motion which carried unanimously. 7. Katie Souder moved to include current and prior pin art and artists names on the website as requested at the AGM. Catherine Comroe seconded the motion which carried unanimously. Brian Duggan agreed to help organize the information. 8. Gary Roush requested committee suggestions by July Brian Duggan moved to approve Letter of Intent for Mississippi Valley and Missouri Rhineland supports at the 2016 specialty with the Mississippi Valley and Missouri Rhineland supports contingent on approval of judges. Wanda Sirianni seconded the motion which carried unanimously. The date for next meeting was scheduled for July 22, 2015 and 8:30 PM ET. The meeting adjourned at 9:57 PM ET. Minutes of September 24, 2015 The meeting was called to order at 8:33 PM Eastern Time. Note: Catherine Comroe left the meeting just prior to the discussion of item Brian Duggan moved to include a response to Jeff Lipp s letter in the NL explaining that the Board Of Directors will take steps to advise judges not to enter any SCOA competitions during the period of the SCOA National. Minutes of October 29, 2015 The meeting was called to order at 8:49 PM Eastern Time. Catherine Comroe moved to approve sponsoring a medallion award to the top winning saluki in agility at the Eukanuba invitational at the cost of $45, subject to donations covering the cost. Specialties to be held on June 23, 2016 and June 24, 2016 in Canby, OR as submitted by Pam Sirianni. Brian Duggan seconded the motion which passed unanimously. 5. Debbie Feliciano moved to approve the proposal for the Saluki Club or America National Specialty to be held June 6-10, 2017 (since changed to June 13-17, 2017) in Gray Summit, MO as submitted by Russell McFadden. Brian Duggan seconded the motion which carried with Catherine Comroe voting against the motion. 7. Brian Duggan moved to approve the nominating committee consisting of Linda Scanlon as chair, Terri Smith, Sue Meier and Kevin Carlson as the 1 rst alternate and Kim Anselmo as the 2 nd alternate. Catherine Comroe seconded the motion. The motion carried with Linda Scanlon abstaining. 8. There was a brief discussion regarding the Breeder Referral Committee. Respectfully submitted, Brian Duggan Archives report Susan Jackson, of the Trophy Maintenance Committee, requested information regarding winners and owners of various trophy winners. With few exceptions, the requested information was located and sent to Susan. Carol Fisher, also of the committee, provided information that had not been in the archives as part of this project. Respectfully, Diana Farmer Book of Records No Book of Records will published in Health and Genetics Committee Grant 01426: Personalized Medicine for the Treatment of Canine Mast Cell Tumors Principal Investigator: Dr. Douglas H Thamm, VMD Research Institution: Colorado State University Grant Amount: $89, Start Date: 1/1/2011 End Date: 6/30/2015 Progress Report: End-Year 4 (FINAL) Report Due: 6/30/2015 Report Received: 6/29/2015 Report to Grant Sponsor from Investigator: While surgery remains the mainstay of treatment for canine mast cell tumors (MCT), surgery alone is not curative in some cases, and not possible in other cases. Medical therapy remains an important component of MCT therapy. New drugs that affect signaling through the KIT growth factor receptor are showing considerable promise for the treatment of canine MCT, and MCT with mutations in the KIT protein that make it constantly active may be more sensitive to KIT inhibitors. The drug combination vinblastine and prednisone has roughly the same effectiveness as KIT inhibition against canine MCT; however, the two treatments have not been compared head-to-head, and it is not clear whether vinblastine or KIT inhibitors are more appropriate for the treatment of MCT without KIT mutations. We recently developed a rapid test, which can be performed on fine-needle aspirates, to

We investigated the predictive value of KIT mutation status using this rapid genotyping assay, as well as KIT staining on biopsy sections, in dogs with measurable MCT randomized to receive either vinblastine or the KIT inhibitor toceranib (Palladia). Randomization utilized a novel adaptive statistical strategy that makes use of the KIT assay results. 87 evaluable dogs were enrolled in this study at 4 participating study sites. 59 dogs were randomized to receive toceranib and 28 dogs were randomized to receive vinblastine. 5 dogs remain on-study. Statistical analysis will take place when these dogs have been removed from study. The results of this study will clarify whether KIT mutation testing is a useful decision-making tool for the selection of the best possible medical therapy for dogs with MCT. Grant 01771: Defining the Unique Genetic Markers in Dogs That Define Immune Function, Disease Resistance and Tissue Transplantation Principal Investigator: Dr. Beverly Torok-Storb, PhD Research Institution: Fred Hutchinson Cancer Research Center Grant Amount: $178, Start Date: 1/1/2013 End Date: 12/31/2015 Progress Report: Mid-Year 3 Report Due: 6/30/2015 Report Received: 6/29/2015 Report to Grant Sponsor from Investigator: The Major Histocompatibility Complex (MHC) is a region of DNA on a chromosome that encodes genes that help control the immune response. The proteins encoded by the genes of the MHC are displayed on the surface of white blood cells (leukocytes) and are referred to as dog leukocyte antigens (DLA). DLA provides cells with the ability to distinguish foreign pathogens such as bacteria and viruses from self. The MHC genes are highly polymorphic i.e., each gene has many different forms or alleles. Matching these MHC alleles between the donor and recipient is important for the success of stem cell and organ transplantation. Specific patterns of MHC alleles are inherited on dog Chromosome 12, one from each parent. The unique pattern of alleles within the MHC of one parental chromosome 12 constitutes a haplotype. Since an entire chromosome 12 is passed on from each parent it is easy to perform DLA-typing to match for haplotypes within a family. However matching haplotypes from unrelated dogs is more challenging since different patterns of alleles may be present in their MHC. In humans, finding an unrelated donor with an MHC match for both haplotypes requires a registry of hundreds of thousands of potential donors, a number predicted by the known diversity of the human MHC. Defining the diversity of the canine MHC will help create strategies for more accurate DLA-typing and thereby improve transplantation results from unrelated donors. Better matching of MHC genes between prospective donor recipient pairs is only one important consequence of better DLA-typing. Importantly, since the MHC controls the immune response, a better understanding of the diversity of MHC genes may help us to discover an association between these genes and various diseases, which in turn should inform mechanistic studies to define disease processes and develop new therapeutics. The goal of the project is to construct haplotypes of DLA alleles from about 1200 dogs of about 50 AKC pure breeds. This can be divided into 4 major tasks. 1. Typing the 4 polymorphic class I genes 2. Typing the 3 polymorphic class II genes 3. Cloning and/or confirming new and ambiguous alleles for all class I and II genes. 4. Analyzing and deriving haplotype compositions. Aim 1, nearest allele identities have been established for two of the 4 class I genes, viz., DLA88, and DLA79 in over 1000 dogs. An unexpectedly large number of new alleles have been found for both of these genes. This has necessitated more work than anticipated to clone and confirm new alleles, Aim 3. Aim 2, Allele identities have been established for all the three class II genes, viz., DRB-1, DQA-1 and DQB-1 in over 1000 dogs. Many new alleles have been found for all three genes. We are requesting a no cost extension of 6 months towards characterizing the large number of new class I alleles we identified. Aim 4, haplotype compositions will be finalized after allele identification is complete. Initial typing of DLA64 and DLA12 genes are complete. Haplotyping

Specific haplotypes have been associated with an increased risk for the development of diabetes and auto immune diseases in humans. Knowledge of these associations has been valuable in understanding disease mechanisms. The distribution and frequency of different forms of each of these genes and their groupings among different breeds will greatly enhance our knowledge of the genetic diversity among breeds. The methodology and data gained from this study will enhance the power of association studies between MHC types and canine diseases. For example, in collaboration with Dr. Leigh Anne Clark s group at the Clemson University in South Carolina, we have identified an association of a specific DLA class I allele with pancreatic acinar atrophy in the German Shepherd Dog. Such a database will also immediately enable tissue transplantation from unrelated but matched donors as a treatment for advanced malignancies and other diseases, among the dogs of most breeds. We have completed the proposed amplification and initial sequencing of all the 7 polymorphic alleles of the 1200 dogs set. However, further detailed characterization is focusing on the two most polymorphic class I (DLA- 88 and DLA-79) genes as we observed an unexpectedly high number of new alleles. Alleles of all three class II polymorphic genes, viz., DRB-1, DQA-1 and DQB-1 were typed in the population of dogs as proposed in the grant. Work is being done in batch mode to confirm new alleles for logistics and cost efficiencies. There is no change in the aims or methodology of the experiments, only in the order of executing methodologically independent steps. Soon after Eukanuba and the holidays we look forward to Meet the Breeds at Westminster in February.
