3 S A L U K I C L U B O f A M E R I C A Page 3 PRESIDENT S MESSAGE Planning is well underway for SCOA s 2015 National Specialty and it looks to be a busy event. It will take place June 2 nd to 6 th 2015 at Purina Farms. As there were last year, there will be supported entries on Saturday, May 30, 2015, at the Mississippi Valley Kennel Club and on Sunday, May 31, 2015, at the Missouri Rhineland Kennel Club. Both will be at the Purina Farms site. I want to thank the Chair, Monica Stoner, and her committees for all their hard work. This issue of the Newsletter contains the Nominating Committee s report on their nominees for 2015 to The list contains nominees for the three open Board member positions. Since the AKC required the revised Bylaws to go into effect when they were approved by the AKC in March 2014, the officers were elected for two-year terms in I want to thank Catherine Comroe and her committee for their work. The Board approved SCOA s first Junior Member at its November Board meeting. I encourage all those who know prospective Junior Members to let them know about the opportunity. I recognize that there is a risk to thank any individuals in specific for their work for SCOA, as we have many members working hard for the club and I can t thank them all individually. But please be aware when you look at the list of committee members and the reports in this Newsletter that there are many members working diligently for SCOA. Sharon Furlong moved to approve the minutes of the July 24, 2014 teleconference as sent on August 19, Katie Souder seconded the motion which carried unanimously. 2. Catherin Comroe moved to approve the Treasurer s July financial reports sent on August 3, Debbie Feliciano seconded the motion which carried unanimously. 3. Catherine Comroe moved to give C-Flash $250 to participate in Meet the Breeds at Eukanuba Kevin Carlson seconded the motion which carried unanimously 4. Debbie Feliciano moved to post supported entry critiques on the website in place of the Newsletter. Jennifer Napier seconded the motion which carried unanimously. 5. A discussion occurred regarding to pledge Parent Club Medallions at Eukanuba at the cost of $ for Conformation, for Obedience and $45.00 for Agility. The discussion showed no interest in the expenditure. 6. Catherine Comroe moved to approve the Junior Membership forms as submitted by Jay Kappmeier. Katie Souder seconded the motion which carried with Jay Kappmeier abstaining. 7. Gary Roush move to appoint Debbie Feliciano as the new chair to the Public Education Committee. Meeting Minutes 09/25/2014 The meeting was called to order at 8:33 PM Eastern time. Jay Kappmeier moved to approve the August financial reports sent on September 1, Debbie Feliciano seconded the motion which carried unanimously. 2. Kevin Carlson moved to approve the Saluki Club of America supports at the Missouri Rhineland Kennel Club and the Mississippi Valley Kennel Club shows held May 31, 2015 and June 1, Brian Duggan seconded the motion which carried with Debbie Feliciano voting no. 3. Katie Souder moved to approve the Saluki League of Southern Arizona Specialty to be held on February 15, 2015 as submitted by Teila Childers. Brian Duggan seconded the motion which carried unanimously. 4. Brian Duggan moved to place the judging critiques in the members section of the website for 30 days from the date of receipt before moving them to the public section. Debbie Feliciano seconded the motion which carried unanimously. 5. Kevin Carlson moved to accept Nathan Lundholm s offer to maintain the SCOA website. Katie Souder seconded the motion which carried unanimously. 6. Jay Kappmeier moved to have Brian Duggan coordinate with the JEC and AKC to submit breed information to AKC to assure all information is accurate and current. Kevin Carlson seconded the motion which carried with Brian Duggan abstaining. 7. Jay Kappmeier moved to require any regional or supports to be held 6 months or less in advance of the National Specialty be approved in advance by the BOD including the judging panel. Nelson s request for saluki exhibitor/breeder information for an article about multi-titled salukis in Dog News. Jennifer Napier will respond. This report contains information on votes to be held in March. A full report will be in the next newsletter, and any matters of significance will be shared with the membership through the Announcements. March is the date of voting for AKC Board of Directors. Other votes will be: Read at the December meeting, to be voted in March: Removing Delegate oversight from Field Trials. This change was brought forward by the Field Trial Committee Read at the December meeting, to be voted in March: Changes the time frame of when the limited registration must be removed for a dog to compete in a breed competition. Currently the Limited designation must be removed before the closing date. The amendment would make it the day of the show. This change was brought forward by AKC Staff. Respectfully submitted, Monica Henderson Stoner, Delegate NATIONAL 2015 The 2015 National will be held at Purina Farms June 2-6 (Tuesday to Saturday) Committees are forming and it s getting exciting. We re planning a health seminar...an update by the doctor who has been doing Saluki heart studies, plus offering dopplers. The reproductive vet will be back for semen collection and storage. On the competition front we re adding a Dual Titled Sweepstakes to recognize owners who have worked with their Salukis in different fields. This is in addition to Obedience, Rally, Lure Coursing, Agility, and of course Conformation. It s not too late to try something new and exciting with your Salukis. You ll be among friends and who will have their own stories to tell. We are again offering Canine Good Citizen tests. CGC is a titling event, and might also help reduce your homeowner s insurance. It s also a great way to test how well your Saluki does listen to you. Make plans now to come to Missouri next June. You ll be glad you did. Committee Reports AKC Gazette Columnist The Saluki column in the AKC Gazette continues to be published in the February, May, August and November issues. There was 1 request for information in the archives which is still being worked on. It should be completed and sent by early December. Respectfully submitted, Diana Farmer, Chair JUDGE SELECTION PROCESS This issue of the newsletter contains the second round of balloting. From the nominations sent we will develop a list of twenty judges to be used for future events, including the 2017 National judge (a foreign judge) and the 2016 National Sweepstakes judge. Respectfully submitted Judge Selection Process Committee Public Education Committee The AKC Meet the Breeds will be held on Saturday, February 14, 2015 in conjunction with Westminster Kennel Club in New York City. I would like to invite everyone who is coming to NYC for Westminster to join in the fun and participate at our booth or at least stop by and spend a few moments with us. The lifetime risk is alarmingly high for some breeds like Golden Retrievers (~20% will die of this disease) and Portuguese Water Dogs (~15% will die of this disease). Furthermore, the risk of hemangiosarcoma is not limited to a single breed. In fact so many dogs are at risk to develop hemangiosarcoma that 40 Breed Clubs designated it as a research priority for Despite considerable efforts to find effective treatments, the outcome for dogs with hemangiosarcoma has changed very little over the past 30 years. We believe this is because our understanding of this disease is still rudimentary, but that is changing. Recent evidence suggests hemangiosarcoma conforms to the cancer stem cell model, where a defined subset of cells is responsible for initiating and maintaining the tumor. These cells are resistant to conventional therapies and they also are very adaptable, being able to survive in a variety of niches. In the case of hemangiosarcoma, the cancer stem cells also retain or acquire the potential to differentiate along several different lineages. For this project, we will use this property against the tumor by modulating factors that support the self-renewal of the stem cell compartment and by inducing their terminal differentiation along alternate pathways that have reduced malignant potential. We propose that disrupting the interactions between hemangiosarcoma cancer stem cells and their microenvironment will enhance the sensitivity of these cells to conventional and targeted therapies and improve the outcomes of dogs with this disease. Grant Objectives: For this project, we will examine the potential to use the multipotency of hemangiosarcoma cells to our advantage by forcing them to differentiate into lineages with reduced malignant potential. 12 Board Reports, Delegate s Report and Committee Reports Page 12 Original Project Description: Lymphoma and hemangiosarcoma are major health problems in golden retrievers, causing both suffering and premature death. As part of our ongoing project, Discovery and Characterization of Heritable and Somatic Cancer Mutations in Golden Retrievers, we have identified several regions of the genome that contain genetic heritable risk factors for lymphoma and hemangiosarcoma in Golden Retrievers. We also identified additional somatic mutations in tumors that occur recurrently in both cancers, some of which are linked to duration of remission when treated with standard of care. Our results indicate that a few heritable genetic risk factors account for as much as 50% of the risk for these cancers. These findings offer the potential to develop tests and strategies for DNA tests that can predict risk for individual dogs, as well as to manage risk across the population as a whole. Indeed, both the inherited risk factors and tumor mutations point to pathways that have been implicated in the pathogenesis of LSA and HSA, and thus should inform the development of targeted therapies. In this proposal we aim to find the precise mutations for the heritable genetic risk factors and to validate markers (mutations) used to determine risk at the heritable loci in a larger independent population of Golden Retrievers from the USA and from Europe in order to develop robust risk prediction tools and an accompanying DNA test. We will identify and characterize tumor mutations and study their relationship to the heritable risk factors, tumor pathogenetic mechanisms, and disease outcome. Report to Grant Sponsor from Investigator: This project has completed the first six months. For analysis of the DNA sequence mutations we identified previously, whole genome sequencing of DNA from several golden retriever blood samples from dogs affected by lymphoma and hemangiosarcoma has been performed. A large number of DNA sequence variants has been detected and are being carefully analyzed. Additional DNA sequencing is underway to provide more information. Collection of blood samples from golden retrievers across the US and Europe has been ongoing and over the coming months we will assess the frequency of the risk haplotypes within these two populations. For evaluation of DNA sequence changes in lymphoma and hemangiosarcoma tumor samples we are compiling cases that will provide sufficient material for multiple analytical approaches. Saluki Health Research, Inc. supported AKC CHF Grants Summary of Cardiac Progress Reports received 6/30/14 Grant 01760: Use of Gene Therapy to Treat Dilated Cardiomyopathy Principal Investigator: Dr. Margaret M. Sleeper, VMD Research Institution: University of Pennsylvania Grant Amount: $146, Start Date: 1/1/2013 End Date: 12/31/2014 Progress Report: Mid-Year 2 Original Project Description: Dilated cardiomyopathy (DCM) is the second most common cause of heart disease in dogs and medical management of the secondary signs is the only therapeutic option. The prognosis for affected dogs depends on the stage of disease and the breed. Doberman pinschers exhibit particularly rapid and uniform progression once congestive heart failure (CHF) has occurred, with most living less than 6 months. Heart function is critically dependent upon calcium dependent signaling. During heart disease, malfunctioning of calcium channels within cardiac cells promotes calcium cycling abnormalities, further inhibiting heart function. Gene transfer strategies to reduce calcium cycling abnormalities ameliorate heart disease in animal models as well as in human clinical trials. We propose a placebo-controlled, double blinded study to evaluate gene delivery approaches for treatment of Doberman pinschers affected with DCM and CHF. Dogs will be treated with intramyocardial injections of saline (control group) or a vector expressing two proteins targeting the myocardial calcium pathway. Medical heart failure management will continue in all dogs and statistical analysis will be performed every 2 months. If a clinical benefit is recognized in the treatment group, the control group will undergo gene delivery. If our results show that the gene delivery to improve myocardial cell calcium handling slows progression of heart failure in Dobermans with DCM, the results will have significant ramifications for this breed as well as the potential to

13 Board Reports, Delegate s Report and Committee Reports Page 13 benefit dogs with many forms of heart disease, since calcium handling proteins are abnormally expressed in dogs with heart disease of varying causes. Grant Objectives: Objective 1: Assess efficacy of therapeutic gene transfer to improve the quality of life of Doberman pinschers with symptomatic DCM using a questionnaire that has been specifically developed and validated for evaluating dogs with cardiac disease. Objective 2: Assess efficacy of therapeutic gene transfer to normalize myocardial structure and dysfunction in DCM using physical examination, echocardiography and electrocardiography. Objective 3: Assess efficacy of therapeutic gene transfer to prolong the life of Doberman pinschers with symptomatic DCM. Report to Grant Sponsor from Investigator: All objectives for the project remain the same but there has been a delay in starting this clinical trial because of some new data, which became available after the initial grant was submitted in late These data showed very successful myocardial transduction using a less invasive approach (the vector was infused into the left ventricle rather than directly injected into the myocardium), however the subjects were puppies rather than adult dogs. We are further investigating this less invasive delivery method in adult dogs to determine whether it will also be effective in adult dogs (the population we plan to treat in this study). Unfortunately the government sequestration and closure in 2013 severely impacted Rob Kotin at the NIH (the collaborator producing vector), which caused significant delays in completing our studies. He then left the NIH and is now working at the University of Massachusetts, causing another delay. However, we have now treated 4 adult dogs confirming that the less invasive delivery technique is effective. We are planning on treating two more within the next month to further optimize the delivery technique and plan to begin advertising the clinical trial in September Our results show we will be able to use this less invasive technique for gene delivery, which will significantly benefit the dogs that are treated and may reduce the study cost. Grant 01994: Early and Accurate Prediction of Mitral Valve Disease Development Principal Investigator: Dr. Sydney N. Moise, DVM Research Institution: Cornell University Grant Amount: $36, Start Date: 1/1/2014 End Date: 12/31/2014 Progress Report: Mid-Year 1 Original Project Description: In the dog, 75% of heart disease is caused by myxomatous mitral valve degeneration (MMVD). Yet between 1960 and 2012 less than 12% of publications concerned MMVD even though it affects ~75% of older small breed dogs. The cause of MMVD likely involves the interplay of genetics, aging, and mechanical insult. A dog s mitral valve opens and closes ~120,000/day under a constant barrage of mechanical forces. With such stress, the struggle of the valvular tissue to stay normal is constant. Our overall hypothesis is that dogs that suffer from MMVD have altered strain profiles on the mitral valve leaflets. This altered strain will be related to the structure of the mitral valve apparatus, which in turn is linked to breed size and/ or chondrodystrophic features of the dog. Using our computer algorithm to assess the motion of the mitral valve leaflets the mechanical signatures of valvular strain can be determined. We believe that these signatures are identifiable at a young age in the breeds most commonly affected with MMVD. Although MMVD may afflict the majority of dogs of some breeds, some individuals are not affected even at an advanced age. We believe such a cohort of aged dogs of high risk breeds have strain patterns matching those of dogs of low risk breeds. A quantitative understanding of mitral leaflet strain will both improve our ability to predict MMVD susceptibility and increase the power and resolution of gene mapping efforts, and if successful will inform new targets and timelines for therapeutic intervention. Grant Objectives: To obtain a quantitative understanding of mitral leaflet strain that will both improve the ability to predict MMVD susceptibility and increase the power and resolution of gene mapping efforts. Report to Grant Sponsor from Investigator: Myxomatous degeneration of the mitral valve (MMVD) is the most common cardiac disease of dogs. Research into the underlying mechanisms of this disorder is uncommon with most studies concerning the treatment of advanced disease. The cause of MMVD likely involves

Likely dogs that suffer from MMVD experience altered mechanical strain on the leaflets of the mitral valve that amplifies the degenerative process. Consequently, we hypothesize that certain mechanical signatures of valvular strain exist in dogs of breeds at risk to develop MMVD and that these signatures are identifiable at a young age. We believe that dogs with abnormal strain patterns will have a mitral valve apparatus that differs structurally from dogs with normal strain patterns and that these profiles of strain will be related to dog size/chondrodystrophic phenotype. Although MMVD may afflict the majority of dogs of some breeds, some individuals are not affected even at an advanced age. We believe such a cohort of aged dogs of high risk breeds have strain patterns matching those of dogs of low risk breeds. We will determine if the morphology of the mitral valve apparatus differs in this cohort. The in vivo leaflet kinematics of the mitral valve will be quantified by computational analysis of image datasets obtained non-invasively by echocardiography. We believe establishing correlations of leaflet strain with structural anatomy and/or breed morphometry will both improve our ability to predict MMVD susceptibility and increase the power and resolution of gene mapping efforts. At this point our progress since the beginning of our grant which was January 16, 2014 has included the acquisition of the quantification of phenotype and echocardiographic examination of a subset of dogs to obtain preliminary data. Due to funding constraints about which the Canine Health Foundation is aware uncertainty about the future has led to the loss of our assistance in the biomedical engineering department. However, Dr. Moise and Dr. Butcher are diligently working together during the next couple of months to identify another individual to assist the cardiologist at Cornell University with this investigation. We are dedicated to the completion and implementation because we believe in the value of this study. Grant 01753: Identification of Genetic Factors That Alter the Severity of Cardiomyopathy Principal Investigator: Dr. Kathryn M Meurs, DVM PhD Research Institution: North Carolina State University Grant Amount: $73, Start Date: 1/1/2013 End Date: 12/31/2014 Progress Report: Mid-Year 2 Original Project Description: Arrhythmic heart disease in the Boxer was first described in the 1980s as Boxer cardiomyopathy but is more commonly called Arrhythmogenic Right Ventricular Cardiomyopathy (ARVC). Recent studies have confirmed that this is an adult onset, familial disease characterized by the presence of ventricular arrhythmias, syncope and sudden death, and is associated with a deletion mutation in the striatin gene in many boxer families. We have demonstrated that boxer dogs with 2 copies of the deletion (homozygous) are most likely to have the more severe form of the disease, however dogs with 1 copy of the mutation are more likely to have variable disease, some will become quite sick while others will remain free of clinical signs. The mechanism for the variability in clinical signs is unknown but is thought to be associated with the concurrent inheritance of other genetic factors. Understanding if these genetic factors exist and what they are will greatly improve the ability to use and interpret the genetic test for the striatin mutation. Grant Objectives: Objective 1: Identify 100, 7 year old boxer dogs that are positive heterozygous for the striatin mutation, 50 with less than 100 VPCs/24 hours (low disease expression) and 50 with at least 500 VPCs/ 24 hours (high disease expression) Objective 2: Perform genome wide association on the 100 samples collected above to identify an association between genes located on specific chromosomal regions and the level of expression of disease Report to Grant Sponsor from Investigator: We have performed complete evaluations on 95 dogs so far and have additional dogs scheduled for evaluation. So far we have identified 44 dogs with high disease expression (at least 500 VPCs day) and only 18 that were positive for the ARVC mutation who had low disease expression (less than 100 VPCs at 7 years of age or older). This suggests that presence of the ARVC mutation is more likely to cause the more severe form of the disease. We have started the initial DNA analysis and have some initial evidence that there may be some genetic regions associated with the more severe form of the disease.

LeVine, DVM, PhD Research Institution: Iowa State University Grant Amount: $51, Start Date: 2/1/2014 End Date: 1/31/2015 Progress Report: Mid-Year 1 Original Project Description: Immune thrombocytopenia (ITP) is a common bleeding disorder in dogs. It occurs when the immune system destroys the body s own platelets, blood cells that prevent hemorrhage. The resulting lack of platelets in some dogs causes mild bruising and in other causes severe, lifethreatening hemorrhage. Veterinarians do not understand what triggers ITP and cannot predict its severity. Consequently, all ITP patients are treated with potent medications that suppress the entire immune system. Many dogs experience treatment side-effects including excessive thirst and urination, ulcers, weight gain, and recurrent infections. For some dogs, the side-effects, rather that ITP, prove fatal. Our first aim investigates the specific causes of ITP. We will identify an ITP disease profile by measuring immune cells and proteins that may be involved in platelet destruction. We will also look for genes associated with the disease in Old English Sheepdogs and Cocker Spaniels, since these breeds are especially prone to ITP. These tests will suggest the specific immune and genetic causes of ITP, so targeted drugs can be developed that suppress just these mechanisms, not the whole immune system. Our second aim is designed to find laboratory markers that predict bleeding severity. Using these markers, veterinarians will be able to reserve aggressive treatment for only those dogs as risk for significant blood loss. Grant Objectives: 1. To investigate the specific causes of immune thrombocytopenia (ITP). 2. To find laboratory markers that predict bleeding severity. Report to Grant Sponsor from Investigator: Our project aim to investigate the specific causes of ITP by identifying an ITP disease profile by measuring immune cells and proteins that may be involved in platelet destruction. We also plan to look for genes associated with the disease in Old English Sheepdogs (OES) and Cocker Spaniels (CS), since these breeds are especially prone to ITP. In this initial study period we have established the infrastructure for a two-pronged attack on canine ITP. 1. Targeted evaluation of breeds at risk: OES and CS. Through the efforts of many dedicated breeders, we have received samples from ITP affected dogs and controls for DNA banking and genetic analyses. We have already found some significant differences in the immune cell profiles of CS compared to other breeds of dogs. We are now exploring these differences to determine whether they make Cockers at particular risk for autoimmune diseases. We are also looking for similar patterns in each new case of ITP as they are enrolled in out study. 2. In-depth immunoprofiling of newly diagnosed ITP cases. We have set up multi-institution collaboration among three veterinary academic and referral centers (Cornell University, Iowa State University, and Cornell University Veterinary Specialists) to enroll active ITP cases. We have developed a novel in-depth immune profile system and a new bleeding severity scoring system. The new bleeding score system is important to standardize grading of each dog s clinical status and relate physical exam findings to laboratory test results. Ultimately, the markers of disease severity that we identify will help veterinarians reserve aggressive treatment for only those ITP affected dogs at risk for significant blood loss. ************************************
