3 MMF Presidents Message 207 Hello Mastiff Lovers. Happy Fall!! This year is flying by. I know there hasn t been a newsletter in a while. We are working out the kinks and trying something new. This newsletter will be the first to be published to our website. We are trying to cut cost with the hard copies. With today s technology we all agreed to upload our newsletter to our website. So bear with us. We also have Pay Pal now on our website. We are still working out those kinks as well. Thank you to all our members for your continued support. Let s look back to 207. We had only one supported show in 207. We Held our Supported Show in Wilmont WI. Hosted by Karen Pettry and Gail Marshack. I would like to thank all of our members for stepping up every time we ask for a trophies sponsor. I m amazed every time how quick they are sponsored. Also a big, Thank You, to all our members that travel to support our shows, without you it wouldn t be a success. MMF also helped Great Lakes Mastiff Rescue in 207. We helped as much as we could at their GLMR picnic. We also made a very generous donation. We printed our new MMF logo shirts and hoodies, designed by Lora Myroup. They turned out amazing. We will be putting in a second order to have some available at the 208 National Specialty. Let s move on to 208. Our first show for this year will be in conjunction with the 208 Nationals. On Monday May 4 th MMF will host a regional Specialty before the start of the Nationals. MMF will be hosting the 208 National Specialty in St Charles IL. May 4-20, 208. Julie Kriebs is our Show Chair, Mark Tichenor is the Co-Chair. Please If you can help in any way please contact me. This is a team effort and we all need to help each other as much as we can. Thank you to all the members that have taken on committees. We always could use volunteers. Let s make this national great as they always are hosted by our club. Our second fall show 208 will be hosted by Lisa Armstrong, site still to be determined. As soon as more info becomes available we will put it on our website. We have had a lot of sadness and loss recently. I want to send our condolences to Sherry Brown and her family for the loss of her Mom, Mary Lou Folkestad. She was a lifetime member of the MMF. To Mary Speer and her family. Mary lost her beautiful Daughter Ashley in an awful accident. We have all watched Ashley grow up in the mastiff ring. To Chris Jones ( Tim Plezbert) and their family for the loss of her Mom Pat Jones. To Jean Portelance and her family for the loss of her husband Dr. James King. Our thoughts and Prayers are with you all and your families. Respectfully Submitted by, Karen Pettry MMF President

11 Condolences Ashley Grace Speer, age 24 of Glenwood City, passed away June 25, 207. Ashley was born May 2, 993, to Mary Lynn (Clanin) and Kemer Speer. Ashleys passion in life was horses, dogs, being an avid reader, and the wild adventures of the outdoors. She enjoyed the activities of hiking, mud runs and warrior dashes, volleyball league, and being in the serenity of nature. Her fearless attitude allowed her to shine in 4-H horse shows and FFA, as well as helping her to achieve national awards in dog shows. Ashley loved being surrounded by her family and friends. Wherever she went, she always carried her infectious smile, great love and sweet compassion for others. Pat Jones of Maple Park, Illinois passed following her battle with terminal cancer. Pat, along with her daughter, Chris Jones (Tim Plezbert), successfully bred beautiful Papillons under the Wingssong prefix for over thirty years. She bred and owned several Papillon Best in Show and Best in Specialty show winners. Pat was often seen dog shows with her daughter Chris, who is a professional handler. Sherry Brown s Mother Mary Lu Folkestad, 80, of Duluth, passed away on Wednesday, Nov. 6, 206 at St. Luke's Hospice with her loving family by her side. Dr. H. James King Passed away peacefully at University Hospital on Monday, May, 207, age 76. 14 Study Reveals Possible Biological Trigger for Canine Bone Cancer February 25, 205 Researchers at the University of Wisconsin School of Veterinary Medicine (SVM) have identified the biological mechanism that may give some cancer cells the ability to form tumors in dogs. The recent study uncovered an association between the increased expression of a particular gene in tumor cells and more aggressive behavior in a form of canine bone cancer. It may also have implications for human cancers by detailing a new pathway for tumor formation. The findings, published online Feb. 3 in the journal Veterinary and Comparative Oncology, may eventually provide oncologists with another target for therapy and improve outcomes for canine patients with the disease. The researchers examined cell lines generated from dogs with osteosarcoma, a common bone cancer that also affects people, with the intent of uncovering why only some cells generate tumors. After the dogs underwent tumor-removal surgery at UW Veterinary Care the SVM s veterinary medical teaching hospital cells from the tumors were grown in the lab. This led to six different cancer cell lines, which were then transplanted into mice. The researchers then looked to see which lines developed tumors and which did not and studied the differences between them. We found several hundred genes that expressed differently between the tumor-forming and nontumor-forming cell lines, says Timothy Stein, an assistant professor of oncology. However, one protein called frizzled-6 was present at levels eight times higher in cells that formed tumors. In the complicated process of gene expression, the genetic information encoded within DNA is eventually converted into RNA and proteins, which are responsible for a variety of vital cellular functions. Frizzled-6 plays a key role in relaying signals from the outside to the inside of a cell, acting as a sort of receiving dock for particular types of information. Molecular connections like this activate pathways, some of which regulate the growth, differentiation and migration of cells when working properly. But when pathways go awry, they may contribute to the development of tumors and tumor-initiating cells. The role of frizzled-6 in this process is not yet fully understood.
15 It s exciting because it s kind of uncharted territory, says Stein, who is also a member of the UW Carbone Cancer Center. While we need more research to know for sure, it s possible that frizzled-6 expression may be inhibiting a particular signaling pathway and contributing to the formation of tumor-initiating cells. The study is a good example of how work at UW Veterinary Care can lead to a better understanding of disease, Stein says, and it highlights how basic science can be a bridge to clinical research. Now I d like to see what this means clinically, he says. Does frizzled-6 serve as a marker of a more aggressive disease? Will it help us improve the accuracy of our prognoses? These are the questions we want to answer. Stein also hopes to continue this line of research in human cancer patients. Meanwhile, the lead author on the study, Lucas Rodrigues, is continuing the investigation in dogs. Now we want to make sure that frizzled-6 is truly what gives these cells the ability to form new tumors, says Rodrigues, a postdoctoral fellow in Stein s lab. While frizzled-6 may be the lone culprit, it is possible that a combination of multiple genes may lead to tumor formation, says Rodrigues. The study was also co-authored by Victoria Thompson, an associate research specialist; Katie Holmes, a 204 graduate of the Doctor of Veterinary Medicine program; and Michael Newton, a professor in the Departments of Statistics and Biostatistics and Medical Informatics.
