November 3, 2011 Dear Janet, Enclosed is Hyphen s Wisdom Panel Insights Mixed Breed Analysis Report for your attention. Our analysis of the DNA of this sample indicates that we have tested the DNA from this dog before with a previous version of the Wisdom Panel. We are therefore alerting this fact to you, and informing you that the results of the second analysis may, in some circumstances, potentially differ from the first analysis we carried out on Hyphen s DNA. The reasons why this can occur are outlined below. First, we periodically make upgrades to the computer program that performs our breed analysis. While the effect of most of these upgrades is minor and are unlikely to affect the amount or nature of the breeds detected in the majority of customer dogs, a few results may come back noticeably different in the amount or variety of breeds detected. The most significant of these upgrades with the potential to change previously tested results was on October 5th, 2009. On this date, we proudly launched the Wisdom Panel Insights test, adding 14 new breeds to our breed library as well as a new product, Wisdom Panel Professional that detects over 200 breeds, subtypes and varieties. Wisdom Panel Insights is now able to detect 171 different breeds present in a mixed-breed dog. New Breeds Added to Wisdom Panel Insights Alaskan Klee Kai Icelandic Sheepdog American Bulldog Irish Red and White Setter Beauceron Miniature Bull Terrier Cesky Terrier Norwegian Buhund Chinook Russell Terrier Dogue de Bordeaux Skye Terrier English Toy Spaniel Swedish Vallhund A second potential reason for observing minor differences between the amounts of a breed detected, or difference in the range of breeds detected in an individual dog tested multiple times by the Wisdom Panel Insights algorithm is driven by the possibility of a small variation in the laboratory analysis. When a sample is tested with our marker panel, occasional run-by-run fluctuations in the number of successfully returned markers can occur. While this is not normally sufficient to alter the range of breeds detected in the analysis, occasionally differences in the breeds detected at our lowest confidence category may be seen. November 3, 2011 Dear Janet, The Wisdom Panel Designer Dog computer algorithms have performed a variety of analyses and Hyphen s results are not consistent with a first-generation F1 cross between a Border Collie and Cocker Spaniel. This is due to the following observed results: In a Principle Component Analysis (PCA), Hyphen s DNA sample does not appear within the expected F1 cluster between Border Collie and Cocker Spaniel. In a PCA, Hyphen s DNA sample does not appear within the expected F1 cluster between Border Collie and Cocker Spaniel when compared to an outgroup. A Wisdom Panel ancestry analysis failed to identify Border Collie and Cocker Spaniel as the sole components of Hyphen s recent ancestry. Using a population genetic analysis program (Structure) to analyse the sample in relation to Border Collie and Cocker Spaniel, the sample had more weighting towards one of the breeds. The observed homozygosity of Hyphen s DNA data is not within the expected range for a first-generation F1 cross between Border Collie and Cocker Spaniel. The following pages contain a detailed summary of the results of each of the separate tests performed on Hyphen s DNA data. BOXER HEIGHT: 21-25 in WEIGHT (SHOW): 55-66 lb WEIGHT (PET): 49-77 lb EARS MUZZLE TAIL The history of the dates back to nineteenth century Germany, where they were used for hunting deer and boar. The ancestors of the include the Bullenbeiszer and the Barenbeiszer, which are now both extinct. The crossing of those breeds with the English Bulldogs of the 1830 s resulted in the as we know it today. s were bred to be hunting dogs and they earned their name from the boxing pose they are known to take when standing on their hind legs. Later in the breed s development, it was made apparent that they were also well-suited for herding and the was used in more than a few circus acts due to its ability to learn tricks quickly and perform them on command. The popularity of s started to increase rapidly in the 1860 s when the German Klub was founded. At the turn of the twentieth century the made its way to the United States and the American Kennel Club recognized the as a breed in 1904. The AKC breed standard for boxers requires that they come only in fawn and brindle (black and brown stripes) with the fawn ranging from a light tan to mahogany and restricts the amount of white seen on the dog. Accepted traits include a black mask commonly seen in the breed. White boxers also have a following; though do not meet the breed standard. BEHAVIORAL TRAITS OF THE BOXER BREED THAT YOU MAY BE FAMILIAR WITH: Intelligent, hard working, and playful dogs, with a high amount of energy. s seem to enjoy dog sports such as agility, flyball, rally and competitive obedience. Eager to learn and respond well to reward-based training using treats and favorite toys. Tendency to jump up on people, sometimes boxing with their front feet when doing so.

AUSTRALIAN KELPIE HEIGHT: 17-20 in WEIGHT (SHOW): 25-45 lb WEIGHT (PET): 25-45 lb EARS MUZZLE TAIL The is a medium sized dog breed that has been around since 1870 in its native land, where over 100,000 members of the breed are found today. They are renowned as excellent herding dogs that are thought to have been predominantly descended from "English Colleys" (Collies) that were imported to Australia during the 19th Century. The name comes from the water described in a book by Robert Louis Stephenson. There is a marked distinction between show and working dogs with different registries existing in Australia for the two types. Working s can be any color and have 3 coat types: smooth, short and rough, whereas show bred s are limited to solid colors only with a short double coat. s are currently recognized by the United Kennel Club in the United States. Working s come in almost any color with 3 types of coat: smooth, rough and short. Some also have a white blaze or points. Show dogs show less variation being solid colors on a short coat only. AKITA HEIGHT: 24-28 in WEIGHT (SHOW): Akita 66-110 lb WEIGHT (PET): 60-121 lb EARS MUZZLE TAIL The Akita is an ancient breed native to Akita city in the Tohoku region of Honshu. The Akita has been a symbol of health and prosperity and bears the status of an imperial symbol and guard. The breed s history began in 1630 when Matagi Inu breeders started crossing their dogs with other breeds indigenous to the Akita region. The resulting breed was known as the Odate Inu, which were crossbred with dogs native to other regions of Japan and larger European dogs, leading to the modern day Akita. In 1931 the Akita was designated as a national monument and became the national dog of Japan. The breed was established as a purebred line at that time. The Akita came to the United States when Helen Keller received one as a gift in 1937. After World War II the breed nearly disappeared as the breed s fur was used for combat clothing and owners tried protecting the dogs by crossbreeding with German Shepherds. Selective breeding began in 1945 to eliminate German Shepherd traits and return pre-war breed characteristics. In 1972 the breed was admitted into the American Kennel Club Studbook. The Akita may be any color including white, brindle or pinto (white with evenly placed patches covering the head and body). Black masks are possible in this breed, but not on white dogs. A white chest blaze is also sometimes seen. BEHAVIORAL TRAITS OF THE AKITA BREED THAT YOU MAY BE FAMILIAR WITH: Intelligent, alert, loyal, and calm dogs. Can be stubborn or willful. Akitas enjoy dog sports such as carting, sledding, agility, tracking, rally and competitive obedience. Stubborn tendencies may be reduced by using motivational tools such as treats and favorite toys in reward-based obedience training. Reputation as a good family dog, but can develop defensive aggression if threatened, scared, or mistreated.

PCA Analysis of Hyphen s DNA sample and samples of Border Collie and Cocker Spaniel from the Wisdom Panel database: Principle Component Analysis (PCA) allows us to see how similar samples are. Closely related samples, like dogs from the same breed, will be expected to be closer together than samples from other breeds. This tends to create a cluster of points for each breed or sub-population within a breed. If a sample is a direct cross between two breeds, it generally lies in-between the two parental breed clusters, and we have created an Expected F1 Cluster to illustrate where we would expect these first-generation F1 crosses to be situated. The PCA below shows Hyphen s DNA data as well as Border Collie and Cocker Spaniel samples from the Wisdom Panel database. Hyphen s DNA sample falls outside the Expected F1 Cluster, where we would expect to find a first generation (F1) cross. This suggests that Hyphen s DNA data is not consistent with a first-generation F1 cross between Border Collie and Cocker Spaniel. Structure analysis: We ran a population analysis software package [Structure: Pritchard Stephens and Donnelly (2000)] on Hyphen s DNA data against Border Collie and Cocker Spaniel. The results of this analysis show what proportion of Hyphen s DNA data clusters with each breed and examining different possible ancestral patterns from the simple first-generation (F1) cross to the more complex ancestries like a second-generation (F2) cross or a backcross. Hyphen s DNA data is closer to one of the breeds in the analysis than we would expect from a first-generation F1 cross of Border Collie and Cocker Spaniel ancestry. Homozygosity Profile: Homozygosity is a measure of how many of Hyphen s genetic markers are identical because both the sire and dam passed down the same marker variant. Purebred dogs tend to have a higher homozygosity than most mixed breed dogs, and first generation F1 crosses generally have a low homozygosity. A higher than expected homozygosity score might indicate a more complex, backcrossed or even purebred ancestry. The observed homozygosity of Hyphen s DNA data does not fall within the range that we would expect for a first generation F1 cross between Border Collie and Cocker Spaniel.
