I. SURVEY OF AMU, AMR IN FRESHWATER FISH CULTURE 1.1. OBJECTIVE: To assess current status of AMU and AMR in tilapia and traditional fishes (freshwater fish) in Nam Sach and Tu Ky districts of Hai Duong province 1.2. ACTIVITIES To investigate status of culture, disease and AMU for freshwater fish production in Nam Sach and Tu Ky districts of Hai Duong province Collect samples for testing of pathogens (Streptococcus spp. & Aeromonas spp.) and test for AMR of the pathogens 1.3. DESIGN Target on farms that have areas of at least 0.2 ha (the average size of farms in Hai Duong province) In each district, select purposely three key aquaculture communes of FW fish Using the principles and techniques of the epidemiological cross-section study and random multi-stage sampling strategy

II. FIELD SURVEY ON AMU AND AMR IN FRESHWATER FISH CULTURE 2.1. Field survey Use random multi-stage sampling strategy in 4 following steps Step 1: Selection of survey location Purposely select 3 freshwater farming communes in each district of Nam Sach and Tu Ky districts from the list that was provided by local veterinary authorities: - Tu Ky district: Tan Ky, An Thanh and Dai Dong communes - Nam Sach district: Quoc Tuan, Thai Tan and Nam Tan communes Step 2: Farm selection List all cultured farms of 6 selected communes and then randomly select 10 farms which have area of more than 0.2 ha from each chosen communes to investigate Farms are coded consistently based on unique number: 2 initials of district name + 2 initials of commune name + order number of farm, eg. TKAT01

NORTH OF VIETNAM A survey will be carried out in Hai Duong province (northern Vietnam): KAP survey Key freshwater fish culture area Nam Sach and Tu Ky districts were selected

II. SURVEY PLAN ON AMU AND AMR IN FRESHWATER FISH CULTURE 2.1. Field survey (cont.) Step 3: Conduct investigation Key technicians of DAH and RIA 1 shall: Provide a training for field investigators before implementing survey Collaborate with leader key technical staff of provincial Sub- DFISH of Hai Duong to visit the selected farms and carry out face-to-face interview to collect relevant information based on the questionnaire

Step 4: Data summary and analysis Collected data will be inputed into MS Access/Excel database Descriptive analyses are carried to summarize the main features and identify variables for statistical analyses Multivariate analyses will be carried out to determinate the odd ratios of AMU and associated factors

III. SAMPLING AND TESTING FOR ANTIMICROBIAL SUSCEPTIBILITY 3.1. Sampling and antimicrobial testing Step 1: During the KAP survey, fish samples will be collected, labeled, preserved and transported to RIA1 lab for detecting the concerned pathogens and testing of AMR Step 2: Testing of samples Bacterial culture methods are used to detect Streptococcus spp. and Aeromonas spp. (these pathogens are the most common in causing diseases in freshwater fish in Vietnam) Use antimicrobial test to assess AMR of the above pathogens to the most common antimicrobials Step 3: Summary of information and data analysis

Testing contents Isolation and identification of Streptococcus caused diseases in tilapia and other freshwater fishes Isolation and identification of Aeromonas caused diseases in tilapia and other freshwater fishes Test for antibiotic resistance to Streptococcus with 14 antibiotics (Neomycin Sulphate; Doxycyline; Hydrochloride; Thiamphenicol flofenicol, Sulfadiazine (SD); Sulfamethoxazole (SMZ/TMP); Sulfadimidine (SM2); Sodium Sulfamonomethoxine (SMM-Na); Enrofloxacin; Flumequine;...) Test for antibiotic resistance to Aeromonas with 14 antibiotics (Neomycin Sulphate; Doxycyline; Hydrochloride; Thiamphenicol flofenicol, Sulfadiazine (SD); Sulfamethoxazole (SMZ/TMP); Sulfadimidine (SM2); Sodium Sulfamonomethoxine (SMM-Na); Enrofloxacin; Flumequine;...)
