3 DOBERMAN PINSCHER The Doberman Pinscher is a relatively new breed, bred around 1890 by Karl Friedrich Louis Doberman, a German tax collector. He aimed to breed a dog that would protect him during his tax collections. Doberman Pinschers are intelligent, loyal, and make for perfect companions as well as guard dogs. The Doberman is a mixture of many different dog breeds that includes Beauceron, German Pinscher, German Shepherd, and Rottweiler. The Doberman is a very athletic dog that often excels in agility courses. Doberman s are trainable and are listed as one of the top five smartest dogs. Fun Fact A Doberman named Cappy saved the lives of 250 U.S. Marines on Guam in 1944 by alerting them when Japanese troops were nearby. 4 MATERNAL LINE Through Jackson's Miss Priss Zandra s mitochondrial DNA we can trace her mother s ancestry back to where dogs and people first became friends. This map helps you visualize the routes that her ancestors took to your home. Their story is described below the map. HAPLOGROUP: C1 Congratulations, C1 is a very exotic female lineage! It is more closely associated with maternal lineages found in wolves, foxes and jackals than with other dog lineages. So it seems dogs in this group have a common male dog ancestor who, many thousands of years ago, mated with a female wolf! This is not a common lineage in any breed, though a good number of German Shepherds and Doberman Pinchers are C1. It is also found in breeds as diverse as Peruvian Inca Orchids and Pekingese; it is rarely found amongst Labrador Retrievers, Border Collies, Siberian Huskies, or Cocker Spaniels. Despite its fascinating origins, it is widely distributed around the globe, and even shows up frequently among Peruvian village dogs. It almost certainly survived at low frequency in Europe for millennia and then was dispersed outside of Europe by colonialism, though not as successfully as some other lineages. HAPLOTYPE: C25 Part of the C1 haplogroup, this haplotype has been spotted in village dogs in Peru and Namibia. Among breeds, it occurs most frequently in Doberman Pinschers, but has also been detected in German Shepherd Dogs and Labrador Retrievers.

6 CLINICAL TRAITS These clinical genetic traits can inform clinical decisions and diagnoses. These traits do not predict a disease state or increased risk for disease. We currently assess one clinical trait: Alanine Aminotransferase Activity. Alanine Aminotransferase Activity result: Low Normal Jackson's Miss Priss Zandra has one copy of a mutation associated with reduced ALT activity as measured on veterinary blood chemistry panels. Please inform your veterinarian that Jackson's Miss Priss Zandra has this genotype, as ALT is often used as an indicator of liver health and Jackson's Miss Priss Zandra is likely to have a lower than average resting ALT activity. As such, an increase in Jackson's Miss Priss Zandra s ALT activity could be evidence of liver damage, even if it is within normal limits by standard ALT reference ranges. More information on Alanine Aminotransferase Activity: Known to be highly expressed in liver cells, activity levels of alanine aminotransferase, or ALT, is a common value on most blood chemistry panels and is known to be a sensitive measure of liver health. Dogs with two ancestral G alleles show "normal" activity. Dogs that have one or two copies of the derived A allele may have lower resting levels of ALT activity, known as "low normal". If your dog's result is "low normal" then when a blood chemistry panel is being interpreted the values that you and your veterinarian consider "normal" may need to be adjusted. Please note that neither a "normal" nor a "low normal" result for this predicts a disease state or increased risk for liver disease. Moreover, this mutation does not associate with increased levels of ALT: If your dog has high ALT levels, please consult your veterinarian.

7 HEALTH Good news! Jackson's Miss Priss Zandra did not test positive for any of the genetic diseases that Embark screens for. 0 AT RISK 1 CARRIER CARRIER CONDITIONS CARRIER status: This indicates the dog has inherited a recessive allele for a genetic trait or mutation. 8 VON WILLEBRAND DISEASE TYPE I (VWF) Carrier VWF GG GA AA GENE NAME CLEAR CARRIER AT RISK Jackson's Miss Priss Zandra is a carrier for a mutation known to cause Type I vwd. We do not recommend breeding Jackson's Miss Priss Zandra to another carrier or a dog of unknown genotype as this could result in the production of affected puppies. DESCRIPTION Coagulopathies represent a broad category of diseases that affect blood clotting, which can lead to symptoms such as easy bruising or bleeding. Dogs with coagulopathies are often at risk for excessive bleeding during surgical procedures; your veterinarian should be informed so that appropriate blood products are at hand in case a transfusion is required. Affected dogs may also require special measures during routine veterinary procedures, and close monitoring during their daily lives. If informed of your dog's condition early, you and your veterinarian can begin taking precautionary measures now. Embark screens for 10 different mutations associated with coagulopathies. These include Hemophilia A and B and von Willebrand Disease, three of the most common inherited coagulopathies in dogs. As well as the precautionary methods described above, an emerging potential treatment for some coagulopathies include protein replacement therapy and gene therapy for a number of coagulopathies. Von Willebrand Disease (vwd) causes easy bruising and excessive bleeding from small cuts and nicks; you may also observe blood in your dog's stool or urine. Affected dogs are also at risk for excessive bleeding during surgery. Von Willebrand Factor is exposed on tissue surfaces upon tissue injury, where it is recognized by platelets and other clotting factors, thus triggering the clotting cascade. vwd is characterized into three types based on clinical severity, serum levels of vwf, and vwf multimer composition. Dogs with Type I vwd have low vwf levels, normal vwf multimer composition, and mild to moderate clinical signs. CITATIONS Brooks et al 2001 (
