1 Name: Block: Muppet Genetics Lab Due: _ Introduction Much is known about the genetics of Sesamus muppetis. Karyotyping reveals that Sesame Street characters have eight chromosomes: three homologous pairs and one pair of sex chromosomes. pair #1 and #2 are the largest and have a centrally placed centromere. pair #3 is much shorter with the centromere at one end. Sex is determined by X and Y chromosomes just as it is in the related species, Homo Sapiens. Through the process of gene mapping, geneticists have found the specific locations of the genes for eye shape (A), nose color (B), fur length (C), body color (D), blood type (E), and hair type (F). By observing the offspring of many crosses, geneticists also have determined the types of inheritance that several of these genes exhibit. In this lab, you will determine the phenotype and genotype of seven Muppet characters, mate two characters, and produce two Muppet offspring. #1 #2 #3 X Y A C D E B F There are five parts: Part 1: Muppet Phenotypes and Genotypes Part 2: Muppet Mates Characteristics Part 3: Punnett Squares for Muppet Offspring Part 4: Muppet Offspring Characteristics Part 5: Muppet Family Portrait 1
2 Requirements Part 1: Muppet Phenotypes and Genotypes marks For each Muppet, identify the phenotype and possible genotype of each Muppet using Table 1.1 and Table 1.2 found on the website Part 2: Muppet Mates Characteristics Choose two Muppets (one male and one female) to mate Identify the phenotype, genotype and type of inheritance for each trait 4 marks Part 3: Punnett Squares for Muppet Offspring 12 marks Using your chosen Muppets, fill in Punnett Squares for each given trait Indicate the probabilities expressed as ratios and percent for each possible trait Part 4: Muppet Offspring Characteristics marks Create two Muppet Offspring (one male, one female) using the results of your Punnett Squares First offspring must have the most likely characteristics (i.e. highest probabilities) Second offspring may have any characteristics Part 5: Muppet Family Portrait marks Detailed drawing of Muppet parents and Muppet offspring Phenotypes of each Muppet should be able to be recognized from the drawing Drawn on.5 x 11 paper This lab is worth 40 marks and will be based on the requirements outlined above. The final information submitted should be neatly written in ink or typed. Handout will be on website if you wish to use a computer. Your lab grade will be based upon the completion of all requirements, neatness and organization. Evaluation rubric is on pages 7-. Rubric has detailed explanations on how marks will be distributed for each part of the lab. 2

4 Part 2: Muppet Mates Characteristics Using a male and female Muppet from the list, record the phenotype and genotypes for each characteristic. These will be Muppet Parents that you will mate to create two Choosing Muppet Parents with different characteristics is preferred! 5 Part 3: Punnett Squares for Muppet Offspring Fill out the Punnett Squares in order to determine the possible traits of your Using the space below each Punnett Square, determine the probability for each phenotype. First, write the probability as a ratio (i.e. 1:4), then as a percent (i.e. 25%). Note: a Punnett Square for sex is not included! 6 Part 4: Muppet Offspring Fill out the two tables of characteristics of two possible offspring from the Punnett Squares. One Muppet Offspring should have the most likely characteristics (i.e. highest probabilities) from your Punnett Squares. The second Muppet Offspring can have any traits that were possible from your Punnett Squares. Muppet Offspring with different characteristics is preferred! Drawings must meet these criteria: Portrait includes drawings of both Muppet Parents and both Muppet Offspring Phenotypes for visible traits (i.e. eye shape, body colour, etc.) should be identifiable from drawings Colour should be used accurately Label each Muppet with their name. Do not label the traits Drawing should be on a separate.5 x 11 paper If someone can look at your drawing and correctly identify the phenotype for each trait for all Muppets, then your drawing is good enough! Marks are awarded for identifying traits, not artistic ability. 6
